A New Weight Loss Pill Could Change the Future of Obesity Treatment

For many people struggling with obesity, weight loss injections such as Wegovy® and Mounjaro® have been game-changing treatments. However, not everyone is comfortable using injections, and access can sometimes be limited due to supply issues and cost.

Now, a new daily tablet called orforglipron is generating excitement after the results of a major international clinical trial showed significant weight loss in people living with obesity.

What is Orforglipron?
Orforglipron is an oral GLP-1 receptor agonist being developed by pharmaceutical company Eli Lilly. Like other GLP-1 medicines, it works by mimicking a natural hormone that helps regulate appetite, slows stomach emptying, and helps people feel fuller for longer.

Unlike current GLP-1 treatments such as Wegovy® (semaglutide) and Mounjaro® (tirzepatide), which are given as injections, orforglipron is taken as a simple once-daily tablet.

What Did the Study Find?
The phase 3 clinical trial involved more than 3,100 adults living with obesity who did not have type 2 diabetes.

Participants received one of three doses of orforglipron (6mg, 12mg, or 36mg) or a placebo tablet over a period of 72 weeks. All participants also received support with healthy eating and physical activity.

The results were encouraging:

Participants taking the highest dose (36mg) lost an average of 11.2% of their body weight.
More than half of participants on the highest dose achieved weight loss of over 10%.
Nearly 1 in 5 participants lost more than 20% of their body weight.
Weight loss outcomes were significantly better than those seen in the placebo group.
Researchers also reported improvements in:

Waist circumference
Blood pressure
Triglyceride levels
Cholesterol levels
These improvements suggest potential benefits not only for weight management but also for overall cardiometabolic health.

What About Side Effects?
As with other GLP-1 medications, the most reported side effects were gastrointestinal symptoms such as:

Nausea
Diarrhoea
Vomiting
Stomach discomfort
Most side effects were mild to moderate and were consistent with what has been seen with other medicines in this class.

Could This Improve Access to Weight Loss Treatment?
One of the most exciting aspects of orforglipron is the possibility of making effective obesity treatment more accessible.

Many patients prefer the convenience of a tablet compared with weekly injections. Oral treatments may also be easier to manufacture, store and distribute, potentially helping to improve availability and reduce costs in the future.

The National Institute for Health and Care Excellence (NICE) has indicated that it will review the evidence for orforglipron as part of its assessment process before any decisions are made regarding NHS use.

Experts have also highlighted that while new medications are welcome, obesity management remains about much more than medication alone. Long-term success is most likely when treatment is combined with lifestyle support, healthy eating, physical activity and ongoing clinical monitoring.

What Does This Mean for Patients?
Although orforglipron is not yet licensed for obesity treatment in the UK, the study results represent another important step forward in obesity medicine.
